Clin Path Bonus Quiz

Lecture 9: Thrombocytes

QuestionAnswer
thrombocytes platelets, stain light purple/blue with dark purple granules, have a lifespan of 5-10 days
megakaryocyte giant, multi-nucleated cell, large amount of cytoplasm
hemostasis the process by which blood is prevented from leaking out of damage blood vessels (CLOTTING)
how does hemostasis occur? platelets will be attracted to the injury, then will stick together as proteins in the blood are activated to form the clot; a platelet plug will form around the torn vessel
normal canine platelet count 250000-500000/uL
thrombocytopenia decreased platelet count, <250000/uL
thrombocytopenia causes increased destruction (Rickettsial infection, immune mediated), decreased production (Aplastic anemia), increased use (hemorrhage), artifact due to clot formation prior to testing
thrombocytopenia clinical signs clinical signs may not be seen immediately due to storage of platelets in the spleen; slow clotting times, unexplained bruising (indicates bleeding subcutaneously)
petechia small pinpoint bruising
ecchymoses large area of bruising
thrombocytosis not usually a clinically significant problem, may be seen in response to trauma or hemorrhage
microplatelets smaller than normal
macroplatelets large or MEGA platelets, usually due to increased production by the bone marrow
infectious granules large granules in the platelets, especially seen in dogs
example of infectious granules Anaplasma platys; a tick-borne bacteria that infects the platelets which leads to their destruction by the immune system
automated platelet count pros rapid results, extremely efficient
automated platelet count cons may be less accurate if platelet clumping has occurred
EPC manual platelet count estimated platelet count; count the platelets in 10 fields on oil immersion, average the platelet count and multiple by 15000 for platelets/uL
DIRECT manual platelet count platelets are counted on hemocytometer; inaccurate due to similar appearance of platelets to debris
feline thrombocytes appearance is similar to canine, but size is more variable; stain light purple/blue, granular appearance, large platelets may be noted in normal feline blood
normal feline platelet count 300000-700000/uL
where do you check for platelet clumps? along the feathered edge
is platelet clumping common in cats? yes, but is more likely due to artifact, not actual differences in cat and dog platelets
