what is the difference between rhodanine method and the rubeanic acid methods rhodanine is more sensitive, but is less specific and may lead to false positives

tech 6-8 (6-10 per sheehan and thicker will stain better)
qc section with copper
reagents SATURATED rhodanine working - binds copper, mayers hematox - stain nuclei, sodium borate - bluing agent
results copper - bright red to red yellow, nuclei - lt blue
what will occur if copper conc. is low fading may occur after coverslipping (therefore copper may be difficult to distinguish from lipofuscin)
what can stop the inherent fading if copper conc. is low use apathy medium for mount
what can also be used as a control fetal liver (in NBF), fixed no longer than 24 hrs berfore paraffin processing.
what occurs if overstaining is done with hematox copper will be masked
what type of reagents should be used analytical grade
since the rhodanine method detects protiens that bind Cu what can occur false positives
