priciple for Von kossa Ca metal substitution/impregnation - Ca salts replaced by silver, uses strong light as a reducing agent
another description of the principle for Von kossa Ca reaction is chemical but is indirect way of Ca detection, silver reacts with carbonate and phosphate of Ca salts. Bright light reduces silver salt to metallic silver
what fixes should be avoided fixes with Calcium, or acidic fixes due to the inherent calcium deposit loss
tech 4-5, chem clean glass, and sunlight preferred (or UV)
qc for Von kossa Ca section with calcium or undecaled bone
reagents silver nitrate 5% - replace Ca salts, soduim thio. 5% - remove unreduced silver, N Fast red - counterstain
results calcium salts - black, background - red
what will also react with this stain melanin
if mercury pigment is present and alcoholic iodine soln' is used to remove the mercury artifact what may occur to the Ca calcium salts will also be removed with alcholic iodine if it is used to remove mercury pigment
what type of light is best and what will occur if alternates are used strong light (quartz halogen better than tungsten lamp), subdued light results in yellow to yellowbrown silver phosphate
what will occur to silver carbonate with this stain it is soluble in sodium thiosulfate and will not be demonstrated
