Principle argyrophil method (ability to bind or be impregnated with silver ions but require reducing agent) - spirochetes can bind silver but need hydroquinone to reduce to visible
fix NBF give best results, DO NOT USE MERCURIAL (zenker, helly, B5) AND CHROMATE FIXATIVES (Zenker, helly, Orth)
QC tissue with spirochets, H pylori, Legionella orgn's, chem cleaned glassware and non metallic foreceps.
micron 4-5
reagents in order for Steiner and Steiner Uranyl nitrate - sensitizer, silver nitrate - impregnation, alcholic gum mastic (K.H. not sure of purpose), reducer/developer (gum mastic, hydroquinone, silver nitrate)
results Spirochetes, legionella, campylobactor - black, background - yellow
if only a few organisms are present what MUST be done with this stain removal of calcium and formalin pigment IS ESSENTIAL
what will occur if xylene substitutes are used they will cause rapid fading of silver stains of microorganisms.
