UND 363 Osmium Tet. Paraffin

principle for osmium tetroxide paraffin proc. osmium tet. combines with fat as a reductioning compound, blackening it. Fat combined with osm. tet. is insoluble in alcohol and xylene and can be processed for paraffin embedding
fix for osmium tetroxide paraffin proc. 10% NBF
tech stain wet tissue block, then process in tissue processor, starting at 70% alcohol. (section should be no thicker than 2mm or osm tet will not penetrate (penetrates slowly)
QC no control need, most tissue contains fat
procedure trim 10% nbf tissue to 2mm, put in osm. tet. (time depends on tissue and size), diff with periodic acid, process routinely starting in 70% alcohol, cut at 4-5 microns, stain with H&E
what can be used as a counterstain for this procedure massons trichrome
results fat - black, other tissue (depends on method)
what amounts of fat can be fixed with this method only small droplets or individual fat cells, gross amounts of fat will not fix
how should facing be done faced carefully, sections taken as soon as possible. osm. tet. doesn't penetrate far and interior of block may not have good preservation or stain well
how will the cytoplasm stain will be grey because of osm. tet.
