MLS 205 Test I

Procedures (Direct Wet Mount, Concentration & Trichrome Staining)

QuestionAnswer
True or False? When preforming a direct wet mount, undiluted Lugol's Iodine is the best stain to identify parasites. False. Lugol's Iodine should be diluted 1:5 with water before staining.
Why are concentrations of little use when observing loose and watery stool specimens? Loose and watery stool specimens typically contain trophs rather than cysts. Concentrations are used to recover cysts and will likely miss trophs.
Why is the amount of sediment critical when preforming a concentration procedure? If there is too little sediment, your chances of recovering parasites is decreased.
List the advantages of the trichrome staining process. 1) Easy 2) Quick 3) Reliable 4) Used on PVA fixed or fresh sample 5) Solutions are stable 6) Polychromatic and easier to read 7) Transparent stain
You receive a rectal swab coated with stool from a 1 year old experiencing watery stools, excessive gas, bloating, loss of appetite, and dehydration. Their medical record notes that they attend a day care center. What parasites should you check for? None. Swabs are unacceptable. Reject it!
