Endospore stain Malachite green & heat, destain with 95% alcohol, counterstain with safranin. Examples Clostridium botulinium, Bacillus anthracis
Acid-Fast stain Tests for thick mycolic acid layer. Stain with carbol-fuschin and heat, destain with acid-alcohol, counterstain methylene blue. Examples: Mycobacterium tuberculosis, Nocardia, Cryptosporidium.
Caseinase Digests Casein enriched agar, positive will have clear halo, Escherichia coli (+) Bacillus subtilis (-)
Amylase Breaks down starch, pour iodine on starch agar, clear means no starch. Escherichia coli (-), Bacillus subtilis (+)
Lipase Breaks Tween80 (polysorbate80) to oleic acid and polyoxyethylated sorbitol. Positive turns broth pink. Mycobacterium smegmatis (+) Escherichia coli (-)
Gelitinase stab bacteria into gelatin medium, positive is if medium liquefies. Escherichia coli (-) Staphylococcus aureus (+)
Dilution factor formula (volume sample)/(Volume sample+volume blank)
CFU/mL formula CFU/(df*volume used of dilution)
