Triple Sugar Iron Test

| What causes black precipitate? | Fe salt in the medium contact with H2S |
| Phenol red? if acid? If alkaline? | pH indicator. Acid: Yellow, Alkaline: Dark Pink. |
| E. coli | Enterobacteriacae, Gram negative bacilli |
| A/A? | acid produce -> G, L or S fermented to acid. |
| K/A | G fermented to acid and protein break down to alkaline end product. |
| K/K | no sugar fermented. or protein converted to alkaline end product aerobically and anaerobically. |
| K/- | No sugar fermented. protein converted to alkaline product aerobically. |
| -/- | Organism fails to grow. |
| Blackening along stab line H2S | H2S produced. |
| Bubble,break in the agar | caused by gas built up. |
