Decarboxylation of amino acid and various substrate

| What is amino acid composed of? | Amino group NH2 and Carboxy acid group COOH |
| what is decarboxylase? | An enzyme, remove COOH, result: alkaline amine and CO2 |
| What is lysine? | Amino acid, if it is decarboxylated, yield to cadaverine. |
| Why use lysine? | Test for lysine decarboxylase activity. |
| What is ornithine? | Substrate, if is decarboxylated, yield putrescine. |
| What kind of media is used for decarboxylase test? | MIO, Lysine |
| What is MIO? | Motility, Indole, Ornithine |
| Why we used mineral oil? | because it will provide an anaerobic environment for glucose fermentation to acid. Acid will activate decarboxylase enzymes. |
| Positive result? | Purple and turbid throughout -> positive -> ornithine is decarboxylated to putrescine. |
| Negative result? | Purple at the top and yellow below -> negative -> Organism lack of ornithine decarboxylase enzyme. Yellow throughout -> Glucose fermented to acid but organism lack decarboxylase enzyme. |
| Purpose of the test? | To detect the ability of an organism to decarboxylase an amino acid. |
