Testing Bacteria

| Catalase Test | Catalase + enzyme destroys hydrogen peroxide |
| Sugar Fermentation Test | inverted tube captures CO2 if it's produced, also indicator (phenol red) turns yellow in presence of acid and magenta in presence of ammonia/base |
| Oxidase Test | Tests for presence of cytochrome oxidase (e- transport chain); turns purple if cytochrome oxidase is present; a strict anaerobe is oxidase - |
| Indole Test | Deamination of tryptophan to indole; media rich in tryptophan; E coli is one of the few indole + enterics |
| KIA/ H2S Production | Thiosulfate reductase catalyzes sulfur to H2S gas, reacts with FeS to form black ppt. KIA has iron/sugars too; bubbles and cracks form if gas is made |
| Urease Test | Catalyzes the breakdown of urea to ammonia and bicarb; pink- basic, yellow otherwise |
| Vogues-Proskauer Test | Positive if pyruvate is fermented into butanediol and CO2 |
| Citrate Test | Tests if citrate can be used as energy source, E. coli is negative and Klebsiella is positive |
