| Question | Answer |
|---|---|
| What is a Spectrophotometry | Analysis of substances based on their ability to absorb specific wavelengths of light. It permits photometric readouts in transmittance, absorbance and concentration. Standard unit is nm=1X10-9m |
| Five steps of Spectrophotometry | 1. Let it warm up for 5-10 min, plug in, turn on select wavelength 2. Label Cuvettes, glass test tubes on top 3. Make a blank 4.Each time change wavelength blank it 5. Wipe down with kim wipes |
| What is a Blank used for | To cancel out any other effects that the other components in the solution may have on the absorbance readings |
| Wavelength where substance has highest absorption | lambda max (h max) |
| How do you read a glassware's volume | Meniscus at eye level |
| What are Transfer pipets? | 1. Transfer Pipet (sim to Pasteur Pipet) Made of plastic has a built in bulb, for dropwise, not accurate. Use for one solution then dispose |
| What are Graduated Pipets? | 2. Graduated Pipet- Specific volumes, divisions imprinted in discrete units, (10th of milliliter or hundreth ml .01, 0 @ barrel, final volume at tip. Must use right size -> Requires Pipet Pump to draw liquid into pipet. |
| What are Volumetric Pipets? | Volumetric Pipets: fixed/variable volumes (0-20 ul, 20-200 ul, 200-1000ul) Range is indicated on it, amount dispensing shown in window. Do not adjust volume or go beyond max. Use disposable plastic tips; remove each after use. To change vol turn dial. |
| How do you use the Volumetric Pipet? | To pick up a liquid: Push to 1st stop in AIR. In water, push fully To drop off liquid: Push to 2nd stop, then remove tip |
