Kirby-Bauer Test Disk diffusion method where bacteria is inoculated over a Mueller-Hinton plate and disks with antibacterial agents are stamped onto plate, zone of inhibition noted
Mueller-Hinton plate Standardized plate with consistent amounts of nutrients and depth. Does not contain PABA which may interfere with antibiotics.
Macfarland turbidity standard Uses turbidity to estimate bacteria concentration.
Staphylococcus morphology colony-pinhead golden round colonies cell- round gram positive in grape clusters
Streptococcus morphology colony-pinpoint round colonies cell-round gram positive in chains or pairs highly pathogenic
hemolysis ability to break down red blood cells, ALPHA-incomplete lysis (greenish halo), BETA- complete lysis and utilization of heme, clear halos.
Mannitol salt agar Selective for salt tolerant bacteria (staphylococcus) and differential for mannitol fermentation (phenol red turns yellow in acid)
GI bacteria. enterics (salmonella e coli), lactobacilli enterococci, clostridium, candida (yeast), protozoa, bacteriosides
lactose fermenters/nonfermenters coliforms can ferment lactose, paracolons cannot
MacConkey agar differential and selective agar containing bile salts and crystal violet, which inhibits gram positives, also contains lactose and neutral red (clear at pH 6.8 and higher)
EMB agar Eosin Methylene Blue Levine agar. Methylene blue and eosin inhibit gram positives and some negatives. lactose fermenters appear blue/black with a metallic sheen if enough acid is produced
SS agar Salmonella shigella agar differentiates those from other enteric bacteria. contains bile salts&brilliant green that inhibit gram(+)& some (-). contains neutral red&lactose. Contains thiosulfate and iron->H2S->ferric citrate creates a black precipitate
