Learning Objectives 1-3

QuestionAnswer

Difference between 3 domains of life? Eukarya have a distinct membrane bound nucleus; Archaea and Bacteria do NOT have a nucleus.
What is the purpose of microscope? To view microorganisms
What are the different parts of the microscope? Objective Lens (10X, 40X, 100X)- above the specimen Ocular Lens- where you view the specimen High power objective - fine adjustment knob Low power objective - course adjustment knob 100X must use immersion oil
How to calculate total magnification? Objective magnification x ocular magnification
Difference between magnification and resolving power? Resolving power is being able to distinguish between two objects as separate entities; Magnification is combination of magnification of ocular and objective lens
Different types of microscopes Dark field- unstained orgs Phase Contrast-internal structures Fluorescence- medical diagnosis Electron Microscope- TEM + SEM
Correct transportation of microscope Hold arm with one hand and base with the other hand
What is Endosymbiotic Theory? Helps explain why prokaryotic characteristics are found in eukaryotic cells
Why are fungi important for decomposition and nutrient cycle? They break down dead and decaying orgs. which is essential to earth's nutrients cycle. They put the nutrients they decompose back into the environment
What is the symbiotic relationship between fungi and plants Fungi provide mineralized nutrients to plants in exchange for continous supply of carbs and other compound
What are structures found in fungi and explain their function? Hyphae used for nutrient absorption Collective mass of hypae are mycelium; seen on foods
Difference in reproduction in mold and yeasts Molds (multicellular) reproduce asexually through spores Yeast (unicellular) reproduce asexually through budding
Discuss dimorphism Means that some fungi have two different growth forms Mold grows at room temp. Yeast grow at human body temp.
Properties of Sabouraud's Dextrose Agar Has high sugar concentration and slightly acidic which makes it unsuitable for bacteria to grow. (used to fungi)
What is lactophenol cotton blue stain? It is used to view fungal specimens because it kills and preserves the organism.
What are the pros and cons of algae? Pro: they produce majority of the world's oxygen (primary producer) Cons: produce toxins that cause problems for humans
Differentiate between groups of parasitic worms? Flatworms: platyhelminthes Roundworms:Nematoda Tapeworms: Cestodes (beef lamb, pork) (most common)
