1.2- Structure and Replication of DNA

QuestionAnswer
What process is used to amplify DNA? PCR
What is the third stage of PCR? DNA is heated to between 70-80 degrees
During PCR, why is DNA heated to between 70-80 degrees? For heat tolerant DNA polymerase to replicate region of DNA
What is the first stage of PCR? DNA is heated to between 92-98 degrees
During PCR, why is DNA heated to between 92-98 degrees? To separate strands
What is the second stage of PCR? DNA is cooled to between 50-65 degrees
During PCR, why is DNA cooled to between 50-65 degrees? To allow primers to bind
What components create DNA Nucleotides? Deoxyribose sugar, phosphate, base
In DNA, what pairs with Adenine? Thymine
The base sequence of DNA forms the Genetic code
What bonds form between bases? Hydrogen
In DNA, what pairs with cytosine? Guanine
What is the primary enzyme needed for DNA replication? DNA polymerase
What does DNA polymerase need to start replication? Primer
What is a primer? A short strand of nucleotides
Where do primers bind? 3’ end of template DNA
What do primers allow to happen? Polymerase to add DNA nucleotides
What needs to happen to form 2 template strands? DNA is unwound and hydrogen bonds between bases are broken
What DNA strand is replicated continuously? Leading
What enzyme is used to join fragments of DNA together? Ligase
What DNA strand is replicated in fragments? Lagging
What is replicated in fragments because DNA polymerase can only add nucleotides in one direction lagging
What strand is replicated in fragments because DNA polymerase adds nucleotides in both direction leading

Use these flashcards to help memorize information. Look at the large card and try to recall what is on the other side. Then click the card to flip it. If you knew the answer, click the green Know box. Otherwise, click the red Don't know box.

When you've placed seven or more cards in the Don't know box, click "retry" to try those cards again.

If you've accidentally put the card in the wrong box, just click on the card to take it out of the box.

You can also use your keyboard to move the cards as follows:

If you are logged in to your account, this website will remember which cards you know and don't know so that they are in the same box the next time you log in.

When you need a break, try one of the other activities listed below the flashcards like Matching, Snowman, or Hungry Bug. Although it may feel like you're playing a game, your brain is still making more connections with the information to help you out.

To see how well you know the information, try the Quiz or Test activity.
