Tricyclic Antidepressants Amitriptyline, Nortriptyline, Block uptake of 5-HT and NA at presynaptic cell. Compete for binding site on carrier protein. Depression, takes 2 weeks. Sedative effect.Chronic pain. Also an agonist of H1 receptors and a1-adrenoceptors. Cost effective, limited by toxicity. Cost effective Wt gain, urinary retention, arrhythmia, hepatotoxicity, blood disease... CI; glaucoma, epilepsy, aft' MI, prostatic hypertrophy.
MAOI (Monoamine Oxidase Inhibitors) Moclobemide, Phenelzine Inhibits monoamine oxidase so red' metabolism of 5-HT, NA, DA Refractive and atypical depression Int' cheese (tyramine-rich food). Risk of hypertensive crisis
Lithium Lithium Unknown Prophylaxis and treatment of bipolar affective disorder. Narrow theraputic index; coma,convulsions, nystagmus wt gain, oedema, tremour, sedation, ataxia, nausea + vomiting
SSRI (Selective Seritonin Reuptake Inhibitor) Fluoxetine, Sertraline, citalopram Monoamine reuptake blocking- prevents reuptake of 5HT (serotonin) by postsynaptic terminals. Depression, takes 6-12 weeks to take effect. Well tolerated. Can lead to seratonin syndrome Vasculitic rash, seizures, anorexia and sexual dysfunction, anorexia, wt loss, hyponatraemia
