DrugMOA & ADRs
Diethylstilbestrol (DES) MOA: estrogen both synthetic & nonsteroidal, no longer in use b/c >ly increases the risk of clear cell cervical or vaginal carcinoma in the daughters of women who took it
Ethinyl estradiol MOA: estrogen synthetic & the strongest, used for contraception, HRT & replacement tx in deficient individuals; ADRs: less of lrg 1st pass b/c synthetic, nausea & vomiting
Estrone MOA: estrogen synthetic prodrug of ethinyl estradiol, used for contraception, HRT & replacement tx in deficient individuals; ADRs: lrg 1st pass-bind to albumin & sex binding globulin, nausea & vomiting
Mestranol MOA: estrogen synthetic prodrug of ethinyl estradiol, used for contraception, HRT & replacement tx in deficient individuals; ADRs: lrg 1st pass-bind to albumin & sex binding globulin, nausea & vomiting
Hydroxyprogesterone MOA: progesterone used for contraception or endometriosis; ADRs: weight gain, depression, hirsutism, LDL:HDL ratio increases, risk for acne significantly increases
Norethindrone MOA MOA: low dose progesterone used for contraception taken each day or endometriosis, endometrial carcinoma, - postpartum lactation, tx dysmenorrhea, control dysfxal uterine blding, prevent further fetal loss in habitual abortion
Norethindrone ADRs ADRs: weight gain, depression, hirsutism, LDL:HDL ratio increases, risk for acne significantly increases, not as effective as combo or estrogen alone, irregular blding
Norgestrel MOA MOA: low dose progesterone used for contraception taken each day or endometriosis, endometrial carcinoma, - postpartum lactation, tx dysmenorrhea, control dysfxal uterine blding, prevent further fetal loss in habitual abortion
Norgestrel ADRs ADRs: weight gain, depression, hirsutism, LDL:HDL ratio increases, risk for acne significantly increases, not as effective as combo or estrogen alone, irregular blding
Combination Pill MOA: HRT & standard PO contraceptive, most common, used for 3 wks, w/ 7 days hiatus of 'fake' pills 2 + menses, effect on the pituitary & hypothalamus, also work on - follicular development & ovulation, progestin makes cervical mucus impenetrable 2 sperm
