Identifying transformed cells

Not all of the host cells will sucessfully take up a recombinant plasmid 3 issues can occur The recombinant plasmid doesnt get inside the cell The plasmid rejoins before the DNA fragment entered The DNA fragment stick to itslef rather than inserting into the plasmid
What do marker genes on the plasmid do They can be used to identify which bacteria successfully took up the recombinant plasmid
What are the 3 Marker genes used Antibiotic resistance genes Genes coding for fluorescent proteins Genes coding for enzymes
Antibiotic resistance marker genes They are resistanant to tetracycline gene and Resistant to the ampicillin gene
Once DNA fragment is inserted Resistance to the tetracycline gene is disrupted by DNA fragment This gene will no longer create a functional protein
Grow bacterial on agar
Transfer the bacterial colonies to a plate with ampicillin antibiotic in the agar
Transfer the bacterial colonies to a plate with tetracycline antibiotic within the agar
Jellyfish contain a gene which codes to create a Green fluorescent protein (GFP) This GFP gene can be inserted into the bacteria plasmid
When a Plasmid with a GFP gene in what happens when DNA fragment inserted DNA fragment inserted in the middle of the GFP gene This disrupts it and prevents GFP production
Enzyme Markers The enzyme lactase can turn a certain substance blue from colourless
The gene for this enzyme is inserted into the plasmid The DNA fragment inserted in the middle of this gene to disrupt it The bacteria are then grown on an agar plate with the colourless substance
The colonies which cannot turn the colourless substance blue contain the recombinant plasmid
When a plasmid has a lactase gene in The DNA fragment is inserted in the middle of the lactase gene. This disrupts it and prevents lactase production
What does a fermenter do A fermenter is used to grow multiple copies of host cell which have been identified as containing the recombinant plasmid
This large cloned population of host cell Can then produce protein coded for by the inserted DNA fragment
