restriction enzyme an enzyme that cuts a DNA strand wherever it finds a specific short sequence of base pairs
gel electrophoresis a method of separating macromolecules by size and charge by using electrical attraction to pull the molecules through a porous gel
PCR Polymerase chain reaction. A technique to amplify DNA by repeatedly cycling through rounds of denaturing DNA, annealing specific primers flanking the site to be amplified, and copying DNA with a polymerase.
CRISPR-Cas9 A gene editing tool that allows DNA sequences to be altered in almost any way desired in a living cell.
transcription The copying of a gene in a cell’s DNA into an RNA transcript.
intron A sequence in a eukaryotic gene that is transcribed but will not be translated. Introns must be spliced out of the raw mRNA transcript before it leaves the nucleus to be translated.
Poly-A tail A sequence of adenine (A) nucleotides added to the end of eukaryotic mRNA transcripts before they leave the nucleus for translation - length of the poly-A tail determines how many times the mRNA can be translated before it is degraded in the cytoplasm
degenerate code A code in which there is not a one-to-one correspondence between code signals and their translations.
tRNA A small RNA molecule that binds to a specific amino acid and to a specific codon in an mRNA molecule. tRNA molecules act as the means for the ribosomes to assemble a polypeptide with the correct amino acids specified in the mRNA code.
operon A set of genes in a prokaryote that are adjacent to each other and that share a common promoter and regulatory mechanisms.
in-situ hybridization A method for detecting gene expression in tissues by binding an RNA probe (labeled with a dye or fluorescent marker) to mRNA in a fixed tissue sample. It allows detection of gene expression in a specific tissue at a specific point in time.
translation The reading of an mRNA code by a ribosome and tRNA molecules and the resulting construction of a polypeptide based on that code.
