Quiz 8 Lab

QuestionAnswer
What 2 enzymes can allow for bacteria to reduce? Why would bacteria use each enzyme to reduce sulfur? 1. Cysteine desulfraae- remoces sulfur from the amino acid and converts to Pyruvate to make ATP 2. Thiosulfate reductas- used to or anaerobic respiration
What do bacreria reduce sulfur to? Hydrogen sulfide fas (H2S)
What does a positiven result for sulfur reduction look like? Medium turns black
What causes the apperacne of the positive result of sulfur reduction? H2S combines with oxidized iron to form a black precipitate
KWhat does a negative result for sulfur reduction look like? No blackening of the medium
What is the reaction fo the enzyme Tryotophanse? Converts the amino acid Tryptophan into Indole, NH3 (ammonia), and pyruvate
What does a positive result for Indole production look like? Indole combines with aKovac's reagent to form a red color
What does it mean for an organism to have motility? Organism can move through the medium
What does a positive result for motility look like? Medium becomes cloudy or hazy
What dopes a negative result for motility look like? Medium becomes cloudy or hazy
What are the result for this SIM test: Hazy media, surface turns red after adding Kovaz's reagent, no blackening, - Hazy = motility - Red = indople Production - No blackening = no sulfur reduction
What are the results for this SIM test: Blackened media that is not hazy, surface reamins yellow after adding Kovac's reagent - Not hazy = No motility - Yellow after Kovac's reagent = No indole production - Blackening = sulfur reduction
What type of bacteria does the TSIA differentiate? - Enteric bacteria
How does the TSIA differentiate these bacteria? - Sulfur reduction - Fermentation of glucose and lactase/sucrose
Where is the aerobic enviornment on the agar slant in TSIA test? - Slant
Where is the anaerobic enviornment on the agar slant? - Deep butt
What willn TSIA results look like for an organisim that only uses gluclose? - Pink slant - Yellow butt
Why does the TSIA result appear pink on the slant when using glucose? - The slant appears pink because glucose is used up quickly in the aerobic slant forcing them to have to use amino acids when glucose is gone. USing amino acid produces ammonia (NH3), causing the phenol red in the medium to turn pink
Why does the TSIA result appear yellow on the butt when using glucose? - The butt is yellow because glucose use is slower in the anaerobic butt so glucose is not fully used up after the incubation period. The acid produed from carb use turns the medium yellow
What will TSIA results look for an organism that used glucose and lactose and/ or sucrose? - Yellow slant - Yellow butt
Why does the TSIA results produce a yellow slant when using glucose and lactose and/ or sucrose? - A yellow slant appears because after glucose is used up, tha bacteria starts using lactose and/ or sucrose for the sugar source. This allows for continued carb use which ends up producing acid in the medium making the slant yellow
Why does the TSIA results produce a yellow butt when using glucose and lactose and/ or sucrose? _ A yellow butt appears becuase there is a slower use of glucose in the anaerobic butt which ends up NOT using up glucose after the incubation period causing it to produce acid turning the medium yellow
What might cause agar seperation or cause the agar to lift off the bottom of the tube in TSIA test? - production of gases from carbohydrates use
What will TSIA resultsl look like for an organism that does not use any of the carbohydrates instead use amino acids? - Pink slant - Red butt
Why does TSIA results show that the slant if pink when only using amino acids? - The slant is pink because of amino acid use in aerobic conditions
Why does TSIA results how that the butt is red when only using amino acids? - The butt is red because the slow metabolism in anaerobic area so no color change is detectanble at the end of incubation period
What will the result of TSIA test look like if an organism reduces sulfur? - Medium will turn black; this is usually seen in the butt
If an organism reuces sulfur, how de we know which carbohydrates it uses in TSIA test? - Looking at the slant color since 1. Pink Slant = GLUCOSE ONLY 2. Yellow Slant = Glucose and at least one other sugar
The TSIA Results shows: Butt is black and slant is pink - Sulfur reduction - ONLy glucose is used
The TSIA Results shows: Butt is black and lactose/ sucrose use - Sulfur reduction - Glucose and lactose/ sucrose use
The TSIA Results shows: Both slant and butt are yellow - No sulfur reduction - Glucose and lactose/ sucrose use
The TSIA Results shows: Butt is yellow and slant is pink - No sulfur reduction - Only glucose is used
The TSIA Results shows: Butt is red and slant is pink - No sulfur reduction - No carb use
