Staining
| Question | Answer |
|---|---|
| Define and give examples of absorption | Tissue becomes penetrated by, or absorbs a dye solution and becomes colored without any change or chemical rxn occuring. |
| Adsorption | accumulation of a stain by the surface of a tissue component. Influenced by oppositely charged ions |
| Mordant | reagent used to link the stain, or dye molecules, to the tissue. Many mordants are metal |
| Basophilic | Acidic substance (anionic, negatively charged) that is easily stainable with basic dyes |
| Acidophilic | Basic substance (Cationic, positively charged) that is easily stainable with acidic dyes |
| Auxochrome | chemical group present in a dye that causes it to bind to certain tissue elements. This group can develop a charge and thus bind to oppositely charged groups present in the tissue |
| Chromophore | the chemical grouping that bestows the property of color on a compound |
| Polychrome | Describes a single dye solution that stains different components different colors by phenomena other than metachromasia |
| Lake | combination of a mordant with a dye. The dye-lake can bind to tissue components and form an insoluble colored deposit. |
| Dye Physiological Properties | -Fluorescent -Metachromatic -Leuko |
| Natural Dyes | CIBOSH Carmine Indigo Brazilian Orcein Saffron Hematoxylin |
| Indigo | From the indigo plant Blue color Can be synthesized artificially |
| Carmine | Very little affinity for tissue usally add metal salt (iron, aluminum) Carmine + aluminum= Mucicarme (stains mucin in goblet cells) |
| Orcein | Boil to create Orcinol Orcinal + air + ammonia=Orcein Brown connective tissue stain |
| Brazilian | Used as a red nuclear stain Brazilwood tree |
| Hematoxylin | Hematoxylin + air=hematin Hematein + metal salt= Biological stain |
| Chromogen | Quinone + Chromophores |
| Dye | Quinone + Chromophore + Auxochrome |
| Leuko Dyes | Dyes that undergo oxidation/reduction to become colorless and then reduction/oxidation to have color return Ex: Schiffs |
| Substantive-Direct | Dye binds directly to tissue |
| Adjective-Indirect | Tissue treated first, then dye is attached |
| Impregnation | Metallic dye deposited onto the structure it is staining, not in |
