Micro Lab Exercise 5

QuestionAnswer
A thin film of bacterial cells placed on a slide to stain is called a? Smear
What must be done to the smear to kill the bacteria? This action also preserves microbes with minimal shrinkage or distortion when stained. It must be fixed
The dry smear is passed through a Bunsen burner flame several times to ________ the bacteria. This may not kill all of the bacteria. Heat fix
Alternatively, the dry smear can be placed on a ______ slide warmer for _____ minutes. 60°C for 5 minutes
If the chromophore is a positive ion, the stain is considered a? Basic Stain
If the chromophore is a negative ion, the stain is considered a? Acidic Stain
Staining procedures that use only one stain are called? Simple Stains

A simple stain that stains the background but leaves the bacteria I stained is a? Negative Stain
What stains can be used to determine cell morphology, size, and arrangement? Simple Stains
How long should you allow the loop to cool for? About 30 seconds
Of what value is a simple stain? To observe morphology and arrangement.
What is the purpose of heat-fixing the smear? If you don't, when you do the wash steps, the specimen will wash off.
And heat-fixing, what would happen if too much heat were applied? The slide could break. Cells could rupture. The cells could burn and become distorted.
Methylene blue can be prepared as a basic stain or acidic stain. How would the pH of the stain affect the staining of bacteria? The higher the pH the more basic it is. It has a positive charge.
