What do typhoid fever, cholera and dysentery have in common? | They are waterborne diseases caused by enteric bacteria (coliforms). |
What is a coliform? | A coliform is a gram negative, falcultative anaerobic, non-spore forming rod that ferments lactose to acid & gas. |
What does the presence of E. coli in water mean? | E. coli in water provides evidence that fecal contamination has occurred. |
What is a presumptive test? | 15 lactose broth tubes with durham tubes are inoculated with water and observed for gas production. Gas production occurs, it is presumed to contain coliforms. |
What does MPN stand for? | MPN stand for most probable number of coliforms per 100 ml of water. |
What is a confirmed test? | Positive cultures from the presumed test are streaked onto selective & differential media to see if the gas producing organisms are gram negative. |
What agar is used in the confirmed test? | Levine EMB agar is used. It includes two dyes, eosin & methylene blue that inhibit the growth of gram positive bacteria. |
What will positive results look like from a confirmed test? | Lactose fermenting bacteria are uniquely pigmented, E coli will appear blue-black with a green metallic sheen. |
What is a completed test? | A Gram stain is done using a representative colony from the confirmed test. |
